Etapa III / 2014. Obtaining and characterization of generation 3 of bioactive injectable materials for bone regeneration – SMART
Abstract In this period, the nanocomposites generation 3 Smart were synthesized, starting from apatite and protein-polysaccharide scaffolds. Compositional optimization and improvement of the obtaining method of the (micro)particles were performed, leading to shape-homogeneous and dimensionally monodisperse materials. complex characterization methods were used to investigate functional properties (such as stability at dehydration-rehydration treatment, stability in relevant biological media), micro-structural features and micro-CT analysis. The comsortium started biomechanical tests. For selected scaffolds, the adherence, proliferation and differentiation of osteoblasts on the surface of mineralized gelatin-alginate microspheres was explored, in addition to the effect of the platelet rich plasma (PRP) on the morphology and cellular functions. Through fluorescence microscopy and scanning electron microscopy it has been shown that MG-63 cells adhere and proliferate onto the microspheres. Morphological changes were recorded during time, consisting in an increase of the contact surface and of the number of filopodia extended onto the surface of the material. A strong effect of the PRP treatment on the initial adherence of cells was noticed, while moderate influence on cell morphology at longer times (7 and 14 days) was also recorded. It has been established that the cells cultured onto the surface of the microspheres respond to the osteogenic induction medium through an increased secretion of osteocalcin. The activity of alkaline phosphatase was more sensitive to the confluence degree of the cells than to the presence of the differentiation inducers. The osteolytic potential of macrophages cultured in the presence of the microspheres was investigated through the level of secreted TNF-alpha and IL-6; a very low potential of induction of the pro-inflammatory citokines in the presence of these materials has been measured. All the activities of the work plan were successfully achieved. All the partners accomplished their tasks.
Etapa III / 2014. Obtinerea si caracterizarea generatiei trei de materiale injectabile bioactive pentru regenerare osoasa – SMART
Rezumatul etapei 2014 In aceasta etapa, au fost sintetizate nanocompozite generatia 3 Smart, pe baza de apatita si matrici proteina-polizaharida. S-au optimizat compozitiile si procedeul de obtinere a (micro)particulelor, obtinand in final materiale omogene ca forma si monodisperse dimensional. Acestea au fost caracterizate prin metode complexe din punct de vedere al unor proprietati functionale (stabilitate la tratament deshidratare-rehidratare, stabilitate in medii biologice relevante) dar si micro-structural precum si prin micro-CT. Au fost demarcate si teste biomecanice. Pentru materiale selectate au fost evaluate aderarea, proliferarea și diferentierea osteoblastelor pe suprafața microsferelor de gelatina - alginat mineralizate precum și influența tratamentului cu plasma bogata în plachete (PRP) asupra morfologiei și functiilor celulare. S-a arătat prin microscopie de fluorescenta și microscopie electronica în scanning ca celulele MG-63 adera și prolifereaza pe suprafața microsferelor suferind în timp modificari morfologice manifestate prin creșterea suprafetei de contact și a numarului de filopodii extinse pe suprafața materialului. A fost observat un efect puternic al tratamentului cu PRP asupra aderarii initiale a celulelor, si modificari moderate la timpi indelungati (7 și 14 zile) ale morfologiei celulare. S-a arătat ca celulele cultivate pe suprafața microsferelor răspund la mediul de inducere osteogenica printr-o secretie crescuta de osteocalcina, și ca activitatea fosfatazei alcaline este mai degrabă dependentă de gradul de confluenta al celulelor decât de prezenta inductorilor de diferentiere. De asemenea a fost evaluat și potențialul osteolitic al macrofagelor cultivate în prezenta microsferelor prin masurarea secretiei de TNF-alpha și IL-6, observandu-se un potențial foarte redus de inducere a citokinelor pro-inflamatorii în prezenta acestor materiale. Prin urmare, toate obiectivele etapei cuprinse in Planul de realizare au fost realizate. Toti partenerii si-au indeplinit sarcinile asumate.
